dev-python/pygtk-2.0.0-r1
and
dev-python/pygtk-2.2.0

If I comment out 

#CFLAGS="-mcpu=7450 -O2 -pipe -fsigned-char -maltivec -mabi=altivec -mpowerpc-gfxopt"
and
#CXXFLAGS="-mcpu=7450 -O2 -pipe -fsigned-char -maltivec -mabi=altivec -mpowerpc-gfxopt"
in make.conf pygtk builds and installs fine.

It compiles fine for me. Can you please provide more information:
- gcc-version (gcc -v)
- a dump of the error it gives you
