I am quite sure blender (as of 2.69) does not use luxrender directly at all, so a USE flag to merely pull in the other package (which isn't even in the tree yet) would be quite pointless.
does one need to rebuild blender with any configuration changes in order to make use of luxrender?

No, both packages are completely independent in respect to compilation. LuxRender exporter for Blender (LuxBlend) is an add-on which can be added to Blender using its built-in add-ons installation function.

Then there will be no useflag for this. We can add it as optional dependency in elog.
