I have a AIO HP 4500 officejet which I tried to installed, after installing cups and hplip I'm missing the ppd file from it although it exists in the tar.

Do you really need the ppd? Newer versions of hplip should be able to set up the printer without a ppd and dynamically create one when you configure the printer. However there are some printers which require a static ppd file in this case you  should build hplip with the static-ppd USE flag. This is all documented in the printing guide [1].

I could not configure my printer using hp-setup without that file.

When I install hplip with the static-ppds USE flag the file is there! Like I mentioned above such cases are documented in the link I gave.
