app-misc/fslint depends on dev-util/glade. Apparently dev-util/glade will no longer work with python 2.7 as emerge now generates USE change message for dev-util/glade: 

The following USE changes are necessary to proceed:
 (see "package.use" in the portage(5) man page for more details)
# required by app-misc/fslint-2.44
# required by @selected
# required by @world (argument)
>=dev-util/glade-3.16.1 -python_single_target_python2_7



Yet for both app-misc/fslint and dev-util/glade, python targets python2.6 and python2.7 are the only permitted targets. i.e.

If I update the USE flag for dev-util/glade as directed, the emerge of dev-util fails for "No supported Python implementation .. "

You can upgrade to 3.16.1 and disable Python 2.6 in the targets, or you downgrade to 3.14.2 and enable Python 2.6 in the targets; this has nothing to do with Python 2.7, which is supported in each version.

Given this, I don't understand why 'emerge' wants me to disable python single target 2.7. Emerge won't continue otherwise.

The title of your bug describes an observation which involves python2_6, but your big is about python2_7; it doesn't tell what the problem is.

That is because app-misc/fslint has no PYTHON_SINGLE_TARGET.
