net-p2p/amule-2.2.2 doesnt use upnp even if the USE flag is enabled because it requires a newer version than is specified in the ebuild.
It must depend on >=net-libs/libupnp-1.6.6 instead of simply net-libs/libupnp.

The configure step alerts about this too:
WARNING! UPnP code has been disabled because libupnp >= 1.6.6 not found (version 1.4.6 is not new enough).
